چکیده

A lthough there are different mechanistic theories for aging, endothelial dysfunction (ED) is a rather neglected player in the aging process. A maladaptive insulin/IGF-1-like signaling (IIS) has a remarkable importance in proaging mechanisms, and insulin has direct effects on ED. Therefore, we assume that the endothelium plays a key role in mediating the aging process in the presence of maladaptive insulin signaling. This latter condition leads to insulin resistance and affects several aspects involved in premature aging, such as body composition, mitochondrial activity, and endocrine function. The present review highlights key mediators and mechanisms responsible for the link between endothelial dysfunction, insulin resistance and aging. In particular, we discuss the sirtuin-1 system, the p66Shc pathway, telomeres, and their interrelationships with endothelial damage and repair.

Hypothesis: Causes of Type 2 Diabetes in Progeroid Werner Syndrome

The premature ageing Werner syndrome (WS) is characterized by the early onset of many age related phenotypes, including graying of hair, cataracts, atherosclerosis, cancer and type 2 diabetes. Type 2 diabetes (DM2) is the loss of blood glucose homeostastis, due to insulin resistance and a failure of acute glucose-stimulated insulin secretion (GSIS) by pancreatic cells. Early compensation for in...

Ultrastructure of the liver microcirculation influences hepatic and systemic insulin activity and provides a mechanism for age‐related insulin resistance

While age-related insulin resistance and hyperinsulinemia are usually considered to be secondary to changes in muscle, the liver also plays a key role in whole-body insulin handling and its role in age-related changes in insulin homeostasis is largely unknown. Here, we show that patent pores called 'fenestrations' are essential for insulin transfer across the liver sinusoidal endothelium and th...

Age-Related Alteration in Serum Adiponectin in Rats and Its Association with Insulin Resistance Markers

Introduction: Adiponectin is one of the most important adipose derived hormone that conflicting data are available about serum changes of adiponectin at different ages. The present study was done to determine the age related changes in serum adiponectin and its association with insulin resistance (IR) indices in aging in male rats. Methods: In this study, serum samples were obtained from male r...

Endothelium‐specific CYP2J2 overexpression attenuates age‐related insulin resistance

Ample evidences demonstrate that cytochrome P450 epoxygenase-derived epoxyeicosatrienoic acids (EETs) exert diverse biological activities, which include potent vasodilatory, anti-inflammatory, and cardiovascular protective effects. In this study, we investigated the effects of endothelium-specific CYP2J2 overexpression on age-related insulin resistance and metabolic dysfunction. Endothelium-spe...

Partial lipodystrophy with severe insulin resistance and adult progeria Werner syndrome

BACKGROUND Laminopathies, due to mutations in LMNA, encoding A type-lamins, can lead to premature ageing and/or lipodystrophic syndromes, showing that these diseases could have close physiopathological relationships. We show here that lipodystrophy and extreme insulin resistance can also reveal the adult progeria Werner syndrome linked to mutations in WRN, encoding a RecQ DNA helicase. METHOD...
